Oito anos depois, quatro start-ups angariaram dezenas de milh\u00f5es de d\u00f3lares, prometendo levar as terapias de tRNA at\u00e9 ao mercado.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<section class=\"elementor-section elementor-top-section elementor-element elementor-element-8ee8570 elementor-section-boxed elementor-section-height-default elementor-section-height-default\" data-id=\"8ee8570\" data-element_type=\"section\">\n\t\t\t\t\t\t<div class=\"elementor-container elementor-column-gap-default\">\n\t\t\t\t\t<div class=\"elementor-column elementor-col-100 elementor-top-column elementor-element elementor-element-f50abbd\" data-id=\"f50abbd\" data-element_type=\"column\">\n\t\t\t<div class=\"elementor-widget-wrap elementor-element-populated\">\n\t\t\t\t\t\t<div class=\"elementor-element elementor-element-00b4f63 elementor-widget elementor-widget-text-editor\" data-id=\"00b4f63\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>O cancro g\u00e1strico difuso heredit\u00e1rio, a beta-talassemia, a distrofia muscular de Duchenne e muitas outras doen\u00e7as t\u00eam em comum o facto de serem causadas por muta\u00e7\u00f5es <em>nonsense<\/em>. Nestas situa\u00e7\u00f5es, uma muta\u00e7\u00e3o pontual cria um cod\u00e3o <em>stop <\/em>num local indevido, interrompendo precocemente a s\u00edntese de uma prote\u00edna, o que leva \u00e0 doen\u00e7a. Mas como pode ser corrigido esse erro?<\/p><p>Uma poss\u00edvel abordagem \u00e9 a administra\u00e7\u00e3o de um tRNA que interprete o cod\u00e3o <em>stop <\/em>como se do cod\u00e3o correto se tratasse, promovendo a continua\u00e7\u00e3o da s\u00edntese proteica. Embora a prova de conceito desta abordagem<sup>1<\/sup> j\u00e1 tenha quase uma d\u00e9cada (e os estudos iniciais datem de 2000<sup>2<\/sup>), atraiu pouco interesse devido a receios sobre a sua seguran\u00e7a. Em particular, existe a possibilidade de se induzir a s\u00edntese de prote\u00ednas t\u00f3xicas, por n\u00e3o interromperem a sua s\u00edntese quando apropriado. No entanto, mais recentemente, mostrou-se que os tRNA terap\u00eauticos induzem apenas um baixo n\u00edvel de cod\u00f5es <em>stop<\/em> ignorados em genes normais.<sup>3<\/sup><\/p><p>A entrega dos \u00e1cidos nucleicos \u00e0s c\u00e9lulas de interesse \u00e9 agora a principal barreira ao desenvolvimento destas terap\u00eauticas, mas avan\u00e7os na nanotecnologia t\u00eam ajudado a super\u00e1-la. Nos \u00faltimos tr\u00eas anos, os grandes volumes de financiamento atra\u00eddos pela Alltrna, Shape Therapeutics, Tevard Biosciences e hC Bioscience \u2013 start-ups de terapias com tRNA \u2013 mostraram que os investidores j\u00e1 n\u00e3o t\u00eam receio desta estrat\u00e9gia.
